Retatrutide dosage is one of the most searched questions about the new triple-agonist weight-loss drugs, and the honest answer is that there is no approved retatrutide dose. Retatrutide is an investigational drug. It has no FDA label, so every dose figure in circulation comes from one of two places: the published clinical trials, or sellers of unregulated "research" product. Retatrutide Dosage in the Phase 2 Obesity Trial
The Doses That Were Studied
The main published source for retatrutide dosing is the phase 2 obesity trial, published in 2023, which randomized adults with obesity, or overweight with weight-related conditions, to once-weekly subcutaneous retatrutide or placebo for 48 weeks. Four target doses were studied: 1 mg, 4 mg, 8 mg and 12 mg once weekly.

How Escalation Worked
Like semaglutide and tirzepatide, retatrutide was not started at the target dose. Higher-dose arms began lower and stepped up at intervals, and the trial also compared different starting doses. The reason is gastrointestinal tolerability: nausea, vomiting and diarrhea in this class are concentrated during escalation, and slower escalation is the standard way to manage them.
Phase 3 and Beyond
A larger phase 3 program (TRIUMPH) is evaluating retatrutide across obesity and related conditions, and began reporting topline results in late 2025; retatrutide nonetheless remains investigational and not FDA-approved. Its doses, escalation schedules and results should be read from the published trial reports themselves. Until an FDA label exists, the phase 3 protocols describe how the drug was studied, not how it should be prescribed.

Most retatrutide dosing charts online are built around research-chemical vials: lyophilized powder of unverified identity and strength, sold "not for human use", with suggested unit conversions. The risks are specific:
- Unknown content. Identity, purity and actual quantity per vial are unverified.
- Unknown sterility. Injectable products made outside regulated facilities carry contamination risk.
- Concentration errors. When the amount in the vial is uncertain, every calculated dose is uncertain too.
- No pharmacovigilance. Adverse events go unreported and untracked.
